Rilpivirine 25 mg

 

Mechanism of Action: 
Rilpivirine inhibits the replication of HIV-1 by binding in a non-competitive manner directly to the reverse transcriptase (RT) enzyme. Like all NNRTIs, rilpivirine binds to the NNRTI binding pocket, a flexible allosteric pocket located at a site adjacent to the DNA polymerizing processing site, resulting in conformational changes and altered function of reverse transcriptase.

Indication: 
Treatment of HIV infection in adults and in children 12 years of age and older who weigh at least 77 lb (35 kg) and who have never taken HIV medicines before, and have a viral load (the amount of HIV in a sample of blood) of 100,000 copies/mL or less at the start of treatment. Rilpivirine is always used in combination with other HIV medicines.

Side effect:
Common side effects
• Abnormal dreams
• Diarrhea
• Fear
• Nausea
• Sleepiness or unusual drowsiness
• Stomach discomfort
• Unusual tiredness or weakness
• Vomiting.
Tell your doctor right away if you have any serious side effects, including: mental/mood changes (such as depression, thoughts of suicide).

Drug Interaction:
Some products that may interact with this drug include:
• Orlistat
• Other HIV NNRTIs (such as efavirenz, nevirapine, delavirdine),
• A certain combination HIV medication (elvitegravir/cobicistat/emtricitabine/tenofovir)
• Proton pump inhibitors (PPIs such as esomeprazole, lansoprazole, omeprazole, pantoprazole, rabeprazole).

 

Pregnancy:
If you are pregnant or breastfeeding, think you may be pregnant or are planning to have a baby, ask your doctor for advice before taking this medicine. Breastfeeding is not recommended during use of this drug; if replacement feeding is not an option, a different drug may be preferred.

Alcohol, Drug and Supplement:
You should take each dose of rilpivirine with a meal. Taking it on an empty stomach may lead to inadequate blood levels and reduced effectiveness of the medication. Grapefruit juice can also increase the absorption of rilpivirine.
